摘要
目的:观察壳聚糖温敏凝胶对体外培养的人牙龈成纤维细胞(GFCs)的生物学作用,为其局部应用于牙周病治疗提供理论依据。方法:采用组织块法分离培养人GFCs,制备50、10、2倍及20%4种浓度的壳聚糖温敏凝胶浸提液,并设立空白对照组,采用MTT法检测其对GFCs增殖状况的影响;碱性磷酸酶(ALP)检测试剂盒检测细胞ALP活性以反映壳聚糖温敏凝胶对GFCs分化状况的影响。结果:在培养初期24h,各浓度浸提液组与对照组比较,GFCs的增殖及ALP活性(A值)差异均无统计学意义(P>0.05);培养48h后,各浓度浸提液组GFCs的增殖及ALP活性与对照组相比差异有统计学意义(P<0.05),其中5倍组对GFCs的增殖及分化状况的影响明显强于25倍组及10%浓度组,差异均有统计学意义(P<0.05)。结论:壳聚糖温敏凝胶有促进GFCs增殖的作用,能增强GFCsALP活性,用于牙周病局部治疗,具有进一步的研究价值。
Objective: To evaluate the biological effects of the chitosan thermosensitive hydrogel on human gingival fibroblast cells (GFCs), and provide information about local application of the chitosan thermosensitive hydrogel on periodontal diseases. Methods: The human gingival fibroblasts were primary cultured through tissue culture method and then cultured in medium containing the chitosan thermosensitive hydrogel at different concentrations.MTT colorimetric assay was used to evaluate the proliferation. And alkaline phosphatase (ALP) kit was used to evaluate the ALP activity of the GFCs. Results:There were no significant differences in proliferation of GFCs and ALP activity at different concentrations of the chitosan thermosensitive hydrogel between the 4 groups and the control group in the early cultures (24 h),(P 0.05). After 48 h of culture, the proliferation of GFCs and ALP activity was significantly different between the different concentration groups and the control group(P 0.05). Conclusion:The results suggest that the chitosan thermosensitive hydrogel could promote the proliferation of the GFCs. Local application of the chitosan thermosensitive hydrogel is feasible for treating periodontal diseases.
