摘要
目的分析宁夏地区牛脑组织博尔纳病病毒(Borna disease virus,BDV)的感染状况,探讨阳性基因扩增片段与德国马源性标准病毒株He/80同源性。方法采用逆转录聚合酶链反应(RT-PCR)、荧光定量巢式逆转录酶聚合酶链反应(FQ-nRT-PCR)分别检测宁夏地区145例健康牛脑组织中BDVp40、BDVp24基因片段,对BDVp24阳性扩增产物进行基因序列测定,并应用BLAST和DNAsist 5.0软件对测序结果进行分析。结果 145例牛脑组织中4例BDVp40、BDVp24均阳性,阳性率为2.759%,且阳性BDVp24片段的核苷酸序列与He/80同源性最高,所编码氨基酸序列相同。结论宁夏地区牛脑组织中可能存在动物源性BDV的自然感染,阳性BDV p24核苷酸序列与He/80具有高度同源性,所编码的氨基酸序列相同。
Objective To investigate the natural infection of BDV in bovine brain tissue in Ningxia,and to explore homology between gene amplification fragments in positive products and the German horse-borne standard strain He/80.MethodsBDVp40 and BDVp24 gene fragments were detected by Revorse Transcriptase Polymerase Chain Reaction and fluorescent quantitative nested reverse transcriptase-polymerase chain reaction(FQ-nRT-PCR) in 145 cases of healthy cattle brain tissue in Ningxia.Gene sequencing was Determined in the positive PCR products and the sequence was analyzed using BLAST and DNAsist 5.0 software.ResultsThere were 4 positive cases in 145 bovine brain tissue samples and the positive rate was 2.759%.The analysis of gene sequence showed BDV p24 in bovine brain tissue from Ningxia was homophylic with strain He/80,and their amino acid sequence remained the same.ConclusionBDV infection may exist in Ningxia bovine brain tissue.BDV p24-positive and Strain He/80 nucleotide sequence are highly homologous.
