摘要
目的:探索艾灸调整实验性类风湿性关节炎(RA)大鼠下丘脑-垂体-肾上腺轴(HPAA)功能的糖皮质激素受体(GR)机制。方法:将SD大鼠随机分为空白对照组、模型组、艾灸组、艾灸加注射RU28362组,用福氏完全佐剂塑造实验性RA病理模型,以GR激动剂RU28362腹腔注射作为干预研究手段,艾灸"肾俞"、"足三里"穴治疗,检测大鼠足容积,用ELISA法测定大鼠血浆CRH、ACTH、GC含量。结果:模型组大鼠足肿胀度明显高于空白对照组(P<0.01),艾灸组大鼠足肿胀度明显低于模型组(P<0.01),艾灸加注射RU28362组足肿胀度与模型组比较无显著性差异(P>0.05);与空白组对照组比较,模型组血浆CRH、GC含量显著增高(P<0.01),ACTH含量显著降低(P<0.05);艾灸组血浆CRH、GC含量明显低于模型组(P<0.01),ACTH含量明显高于模型组(P<0.05),但艾灸加注射RU28362组血浆CRH、ACTH、GC含量与模型组比较无显著性差异(P>0.05)。结论:艾灸对实验性RA具有明显的抗炎效应;艾灸对实验性RA大鼠的HPAA功能紊乱有良性调整作用;海马GR在艾灸调整实验性RA大鼠HPAA功能中具有重要作用。
Objective:To explore glucocorticoid receptor(GR) mechanism of moxibustion adjusting the rheumatoid arthritis(RA) rats' hypothalamus -pituitary -adrenal axis (HPAA) function. Methods:Sprague-Dawley(SD) rats were randomly divided into normal control group,model group,moxibustion group,moxibustion and injecting RU 28362 group(In. group). To produce RA animal models by Freund's Complete Adjuvant (FCA),to inject GR agonist RU28362 as a means of intervention,to moxibust in "Shenshu(BL23)" and "Zusanli (ST36)" acupoints. To detect their feet capacity,and to detect corticotropin-releasing hormone (CRH),adrenocorticotropic hormone (ACTH),glucocorticoid (GC) content in the plasma by enzyme-linked immunosorbent assay (ELISA). Results:Model group rats' feet swelling were significantly higher than control group (P〈0.01),moxibustion group rats' feet swelling were significantly lower than model group (P〈0.01). but there were no significant difference between model group and In. group (P〈0.05). Compared with control group,model group's CRH and GC content in plasma were significantly higher (P〈0.01),while ACTH content were significantly lower (P〈0.05). CRH and GC content of moxibustion group were significantly lower than that of model group(P〈0.01),while ACTH content of moxibustion group was significantly higher than that of model group(P〈0.05),but there were no significant difference between model group and In. group (P〈0.05).Conclusion:Moxibustion for experimental RA model rats' has anti-inflammatory swelling. Moxibustion has the positive role in adjusting for RA's HPAA dysfunction. GR in hippocampus plays the important role of moxibustion regulating RA's HPAA functional.
