摘要
为观察一种改进后的细胞冷冻保护液在－８０℃对小儿造血干细胞的保护作用，将８例未分离的小样本小儿全血骨髓在４℃分别与此细胞冷冻保护液混合，不经程控降温而直接在－８０℃低温保存。在１周、１月和３月时取出，４０℃水浴解冻，检测细胞浓度，同时用０．５％台盼蓝检测细胞存活率。结果表明，该方法在１周、１月、３月后均有很高的细胞回收率和存活率，且无明显差异（Ｐ＞０．０５）。我们认为：该冷冻保护液对小儿造血干细胞有良好的保护作用。该方法的确是一种简便而安全的体外冷冻保存方法。
To observe the efficiency of a modified cryopretective solution for cell pretection at 80C. 8 small volume unseparated childish whole bone marrow samples were mixed with thisnew solution respectively and stored at -80℃ directly without rate-controlled freezing. Theywere detected the cell recovery and trypan blue viability storeed after 1 week I month and 3months. Results show that both cell recover and Trypan blue viability were high and similar(P>0. 05) after 1 week, 1 month and 3 months. It proved that the new cryoprevative solution has a good efficiency to store the children hematopoietic stem cell and the new method invitrois really simple and safe.
