摘要
建立了一种双手性拆分试剂(羟丙基-β-环糊精和羧甲基-β-环糊精)拆分烯丙洛尔对映体的毛细管电泳方法。烯丙洛尔对映体在pH 3.8的80 mmol/L Tris-H3PO4+15 mmol/L羟丙基-β-环糊精(HP--βCD)+5.0 g/L羧甲基-β-环糊精(CM--βCD)条件下获得成功分离。以(1R,2R)-2-氨基-1-苯基-1,3-丙二醇为内标,采用内标法定量,烯丙洛尔对映体在0.25-50 mg/L范围内具有良好线性关系,检出限(S/N=3)为0.25 mg/L,日内、日间RSD均不大于6%,平均回收率为97%-102%。该方法可用于鼠体内烯丙洛尔的药代动力学研究。
A capillary electrophoresis method was developed for the chiral separation of alprenolol using dual cyclodextrins(CDs) of hydroxypropyl-β-CD(HP-β-CD)and carboxymethyl-β-CD(CM-β-CD) as chiral selectors.The optimal separation conditions were optimized.A good baseline was obtained by using a running buffer of 80 mmol/L Tris-H3PO4 containing 15 mmol/L HP-β-CD and 5.0 g/L CM-β-CD at pH 3.8.The quantitative analysis of alprenolol enantiomer was performed by using(1R,2R)-2-amino-1-phenyl-1,3-propanediol(Pro)as an internal standard.Under the optimal conditions,the calibration curves were linear in the range of 0.25-50 mg/L for alprenolol enantiomer and the detection limit(S/N=3) were both 0.25 mg/L.Intra-and inter-RSDs were not more than 6%.Mean recoveries were in the range of 97%-102%.The proposed method could be used in pharmacokinetics studies.
