摘要
目的建立LC-MS法测定人血浆中罗红霉素的浓度。方法色谱柱:LichrospherC18(dp5μm,250mm×4.6mmID)流动相:甲醇-10mmol/L醋酸铵水溶液(含1%的冰醋酸)(80:20,v/v);检测波长为210nm,柱温:30℃;流速:1.0ml/min,取上清液直接进样,进样量为20μl。结果罗红霉素线性范围为0.00989~14.81μg/ml。罗红霉素的最低检测限为0.00989μg/ml,日内、日间RSD均<5%,相对回收率为97.6%~104.5%,提取回收率均>91.3%。结论方法灵敏,简便,可重复的,可用于罗红霉素的血药浓度测定。
Objective To establish LC-MS method for determination Cefepime in plasmal. Methods Lichrospher C18 was used with the mobile phase of methanol/10 inmol/L ammonium acetate solution(containing 1% acetic acid) ( 80/20 ,v/v) at the detection wavelength of 210 nm. The flow rate was 1.0 ml/min. Results Under the condition of this Roxithromycin in plasma was well separated, The calibration curve was linear in the range of 0. 009 89 - 14. 81 μg/ml. The limit of quantification was 0.02 μg/ml. The RSD of intra-and inter day were all less than 5%, the relative recovery was 97. 6% - 104. 5% and the extraction recovery was above 91.3%. Conclusion This validated method is sensitive,simple and repeatable enough to be used in pharmacokinetic studies.
