维脑康胶囊对局灶性脑缺血大鼠脑微透析液中两种氨基酸水平的影响 被引量:8

Effect of Wei Nao Kang Capsule on glutamate and GABA levels in rats' brain:a microdialysis study

摘要 目的:研究维脑康胶囊(WNK)对局灶性脑缺血大鼠脑微透析液中两种氨基酸神经递质水平的影响,探讨维脑康胶囊起到脑保护作用的可能途径。方法:SD大鼠分为大脑中动脉阻塞(MCAO)模型组(n=6)和维脑康胶囊治疗组(n=6),治疗组大鼠灌胃给予维脑康胶囊(150mg/kg),每天两次,连续7天,模型组大鼠灌胃给予等体积蒸馏水;药后第七天制备MCAO模型,同时进行微透析取样。微透析样品经柱前衍生化后,以HPLC-ECD系统分析,柱切换法同时测定谷氨酸(Glu)与-氨基丁酸(GABA)的水平,比较两组大鼠在造模前后微透析液中两种氨基酸含量变化情况。结果:给予维脑康胶囊的大鼠在造成MCAO模型后,脑微透析液中Glu与GABA的增加量比模型组均显著减少(P<0.05)。结论:维脑康胶囊能抑制大鼠局灶性脑缺血发生后脑细胞间液中急剧升高的两种氨基酸的释放;提示维脑康胶囊可能通过下调过量的氨基酸释放而起到脑保护作用。 Objective:To study the regulatory effect of Wei Nao Kang Capsule(WNK)on Glutamate(Glu)and-amino butyric acid(GABA)level in middle cerebral artery occlusion(MCAO)rats' brain microdialysis samples.Methods:Sprague-Dawley rats were randomly divided into two groups,one is MCAO model rats suffered from focal cerebral ischemia(n=6),and the other is WNK treated MCAO rats(n=6).WNK was administrated twice each day at a dose of 150mg/kg ig.while distilled water with same volume was administrated to model rats for 7 days.On the day 7,brain microdialysis samples were collected and during the collection,rats were subjected to MCAO operation,and rats without neurological deficit were excluded.Dialysate were pre-column derivatised and analyzed by HPLC-ECD system,a column-shifting assay was employed to simultaneously determine Glu and GABA.Results:Glu and GABA levels in brain dialysates of WNK treated group are significantly lower than that of the model rats.Conclusion:WNK down regulate both Glu and GABA levels in microdialysis samples which rise dramatically after the rats subjected to MCAO,the data suggests,maybe,WNK protect focal ischemic brain from down regulating over-released amino acid.
