摘要
目的探讨EphA2和E-cadherin在乳腺癌中的表达并分析两者表达与预后的关系。方法采用免疫组织化学SP法检测EphA2和E-cadherin在90例乳腺癌和20例乳腺良性病变组织中的表达,分析两者的表达情况与疾病的临床病理特征之间的关系及两种蛋白间的相关性。并分析90例乳腺癌患者的随访资料,判断EphA2和E-cadherin的表达与患者预后的关系。结果EphA2与E-cadherin在乳腺癌中阳性表达率分别为83.3%和35.6%,两者表达均与与肿瘤大小无关(P>0.05);EphA2蛋白的阳性表达与组织学分级,临床分期,有无淋巴结转移有关(P<0.05),而与病理类型无关(P>0.05);E-cadherin蛋白的阳性表达与病理类型,组织学分级,临床分期,有无淋巴结转移有关(P<0.05)。乳腺癌中EphA2和E-cadherin的蛋白表达呈负相关(r=-0.291,P<0.05)。K-M法单因素分析显示EphA2蛋白的阳性表达患者5年生存率低于阴性患者(P<0.05);E-cadherin蛋白的阳性表达患者5年生存率高于阴性患者(P<0.05)。结论E-phA2和E-cadherin在乳腺癌中分别高表达和低表达,且均与乳腺癌预后密切相关,对判断乳腺癌的预后有重要预测意义。
Objective To study the expressions of Eph tyrosine kinase A2(EphA2) and E-cadherin in breast cancer and their relationship with prognosis. Methods The expressions of EphA2 and E-cadherin proteins were studied in 90 breast cancer samples and 20 benign breast lesion samples by SP immunohistochemical(IHC) analysis to study the relationship of the expressions with clinicopathological characteristics,and to understand the correlation between the expression of EphA2 and that of E-cadherin in breast cancer. The follow-up of the 90 cancer patients was analyzed for the relationship between EphA2 and E-cadherin expressions and prognosis.Results The positive rates for EphA2 and E-cadherin expressions were 83.3% and 35.6% respectively in breast cancer tissues, which were related with histological grade,clinical stage and lymph node metastasis (P〈0.05),but not with tumor size and pathological type (P〉0.05). EphA2 and E-cadherin protein expressions in breast cancer were negatively correlated (r=-0.291,P〈0.05 );K-M univariate survival analysis suggested that the 5-year survival rate was lower in patients with positive expression of EphA2 protein than in those with negative expression(P〈0.05); and higher in patients with positive expression of E-cadherin protein than in those with negative erpression(P〈0.05).Conclusion The expressions of EphA2 and E-cadherin are respectively high and low in breast cancer, and both are closely related to prognosis.Thus they are significant in predicting the prognosis of breast cancer.
