SLE患者外周血单个核细胞NF-κBp65的表达和活化

摘要 目的:确定系统性红斑狼疮(SLE)患者外周血单个核细胞NF-κBp65的表达量和活化量与免疫学指标的相关性和评价糖皮质激素对NF-κBp65表达和活化的影响。方法:分别从SLE患者和正常人外周血提取单个核细胞,利用免疫细胞化学的方法检测NF-κBp65的表达量及活化量。结果:NF-κBp65在活动期SLE患者外周血单个核细胞浆和细胞核内均有表达,而在非活动期SLE病人和正常人外周血单个核细胞仅胞浆内少量表达,细胞核内不表达。NF-κBp65表达量和活化量与血清补体C3和C4水平呈负相关,与抗dsDNA抗体和抗Sm抗体阳性率及SLE病情分级呈正相关。糖皮质激素治疗敏感组NF-κBp65的表达量和活化量低于糖皮质激素抵抗组(P<0.05)。结论:活动期SLE患者外周血单个核细胞NF-κBp65表达量和活化量均与病情分级呈正相关;糖皮质激素可以降低NF-κBp65的表达和活化。 Objective: To determine the expression and activation of NF-κBp65 in peripheral blood mononuclear cells in patients with systemic lupus erythematosns (SLE), and to evaluate the expression and activation of NF-κBp65 after treatment with cortieosteroid. Method: After mononuclear cells were extracted from blood of healthy adult and SLE patients, the expression and activation of NF-κBp65 were detected by immunoeytoehemistry method. Results: The expression of NF-κBp65 was observed in both cytoplasm and nucleus in active SLE patients, while only little expression in cytoplasm in inactive SLE patients and healthy controls. The expression and activation of NF-κBp65 were positively correlated with the positivity rates of anti - dsDNA antibody and anti - Sm antibody, and SLE disease classification. There was a negative correlation with complement C3, CA, The expression and activation of NF-κBp65 were lower in glueoeortieoid - sensitive group than in glueoeorticoid - resistant group ( P 〈 0.05 ). Conclusion: The expression and activation of NF-κBp65 have a positive correlation with the activity of the disease. Glueoeortieoid can reduce the expression and activation of NF-κBp65.
