抑制核转录因子-κB对纤维化肝组织中CTGF表达的影响 被引量:2

摘要 目的探讨纤维化肝脏组织中结缔组织生长因子(CTGF)表达的上调是否与激活核转录因子-κB(NF-κB)有关。方法雌性Wistar大鼠70只,随机分为正常对照组(n=6)、纤维化模型组(n=32)和NF-κB抑制剂吡咯烷二硫代氨基甲酸盐(PDTC)组(n=32)。纤维化模型组和PDTC组各自再随机分为1周、2周、4周、6周组。纤维化模型组给予CCl4建立化学性肝损伤模型;PDTC组大鼠在给予CCl4的同时给予PDTC灌胃。常规HE染色观察肝脏病变,免疫组化法测定NF-κB的表达,Western blot检测CTGF的表达。结果纤维化模型组NF-κBp65、CTGF表达呈递增趋势。与正常对照组比较,模型组肝组织NF-κBp65和CTGF表达明显升高,且两者呈正相关(r=0.875,P<0.01);PDTC组NF-κBp65和CTGF表达显著低于同期模型组,且二者呈正相关(r=0.765,P<0.01)。结论在肝纤维化过程中可能通过激活NF-κB通路上调CTGF的表达。 Objective To explore the effect of nuclear factor κB (NF-κB) on connective tissue growth factor(CTGF) in fibrosis liver. Methods Seventy female Wistar rats were randomly divided into normal control group(n=6),hepatic fibrosis model group(n=32) and pyrrolidine-1-dithiocarboxylic acid ammonium sail (PDTC) group(n=32).Carbon tetrachloride was injected for establishing the hepatic fibrosis model in hepatic fibrosis group.The rats in PDTC group were subcutaneously injected with carbon tetrachloride and given oral administration of PDTC.The pathological hepatic changes were observed by HE.The expression of NF-κB p65 was determined by immunohistochemistry.The expression of CTGF was measured by Western blot. Results The expression of NF-κB p65 and CTGF increased with the administration time.Compared with control group,the expression of NF-κB p65 and CTGF increased significantly in hepatic fibrosis group.The expression of NF-κB was positively correlated with CTGF in hepatic fibrosis group(r=0.875,P〈0.01). In PDTC group,the expression of NF-κB and CTGF was lower than that in hepatic fibrosis group. Furthermore,the expression of NF-κB was positively correlated with CTGF(r=0.765,P〈0.01). Conclusion Activation of NF-κB may up-regulate the expression of CTGF during the development of hepatic fibrosis.
