利用五谷虫蛹壳制备甲壳素和壳聚糖条件的优化 被引量:1

摘要 以五谷虫蛹壳为原料,采用酸碱法提取甲壳素并制备壳聚糖。结果表明,从五谷虫蛹壳中提取甲壳素的最佳工艺为:盐敏脱矿物质的质量分数为5%,处理温度为30℃,处理时间为20h;NaOH脱有机物质的质量分数为5%,处理温度为75℃,处理时间为6h。在此工艺下所得甲壳素产品为白色片状固体,其产品质量指标达到食品级甲壳素标准,其水分含量为6.89%,灰分为0.81%,含氮量为6.51%,提取率为32.3%。壳聚糖制备的最佳工艺为:NaOH质量分数55%,浸泡时间6h,浸泡温度100℃。所得壳聚糖产品为白色片状固体,其水分含量为3.17%,灰分为0.67%,脱乙酰度为80.94%,黏度为20mPa·s,提取率为19.4%。 Maggot was used as raw material to extract chitin and chitosan with the Acid-Alkali method.The optimal techniques for preparation of chitin were as the follows: the maggot was treated with 5% HCl at 30℃ for 20h to demineralize;and then treated with 5% NaOH at 75℃ for 6h to remove the organic substances.The obtained chitin under the conditions was white sheet solid,and its water content was 6.89% and ash content was 0.81%.The nitrogen content was 6.51%,and extraction ratio was 32.3%,and its quality reached to the criterion of food grade.The optimal protocol for preparation of chitosan was as the follows: the chitin was extracted with 55% NaOH at 100℃ for 6h.The obtained chitosan under the conditions was white sheet solid,and its water content was 3.17% and ash content was 0.67%.The degree of deacetylation of the product was 80.94%,and its viscosity was 20mPa.s,and extraction ratio was 19.4%.
