摘要
目的观察异氟醚(isoflurane,ISO)后处理对盲肠结扎穿孔术(cecal ligation and puncture,CLP)后大鼠急性肺损伤(acute lung injury,ALI)的保护作用。方法32只SD雄性大鼠随机分为4组(每组n=8):正常组(N组),未行任何处理;假手术组(Sham组),单纯行开腹关腹术;盲肠结扎穿孔术组(CLP组),行CLP建立大鼠ALI模型;异氟醚组(ISO组),建立CLP模型后4 h腹腔注射液态异氟醚2 ml/kg。术后12 h各组动物腹主动脉放血处死,右肺进行病理切片和肺湿/干比(W/D)测定,组织匀浆测超氧化物歧化酶(SOD)、丙二醛(MDA)和髓过氧化物酶(MPO)的含量;左肺行支气管肺泡灌洗,取上清液测总蛋白、肿瘤坏死因子α(TNF-α)和白细胞介素10(IL-10)的含量。结果肺组织病理学检测结果显示CLP组大鼠肺组织损伤严重,ISO组肺组织损伤较轻。与N组比较,CLP组和ISO组中SOD含量降低(P<0.01),其余检测指标均增加(P<0.01)。与CLP组比较,ISO组SOD和IL-10含量较高(P<0.01);其余检测指标均低于CLP组(P<0.05或P<0.01)。结论在本实验条件下,大鼠腹腔注射液态ISO可发挥抗炎、抗氧化作用,对CLP所致大鼠ALI有部分保护作用。
Objective To investigate the protective effects of isoflurane(ISO) post-treatment on acute lung injury(ALI) induced by cecal ligation and puncture(CLP) in rats.Methods 32 male Sprague-Dawley(SD) rats were randomly divided into 4 groups(n=8 each): normal(N) group,with no treatment;Sham-operated control(Sham) group,with mere incision and closure of the abdomen;cecal ligation and puncture(CLP) group,established as rat model of induced ALI;and isoflurane(ISO) group,with intraperitoneal injection of 2 ml/kg ISO 4 h after CLP.The animals were sacrificed by exsanguination via the abdominal aorta 12 h after CLP.The right lungs were removed for pathological dissection;determination of lung wet-to-dry weight ratio(W/D);determination of the content of superoxide dismutase(SOD),malondialdehyde(MDA) and myeloperoxidase(MPO) in the pulmonary homogenate.Following lavages of the left lungs,the supernatant from the bronchoalveolar lavage fluid(BALF) was determined for the total protein level,the content of tumor necrosis factor-α(TNF-α) and interleukin-10(IL-10).Results The pathological results showed that rats in CLP group had serious lung injury,while the rats in ISO group had slight injury.In both CLP and ISO groups,the SOD content decreased(P0.01) and the other parameters increased(P0.01),as compared to the N group.The SOD and IL-10 contents were higher in ISO group than in CLP group(P0.01).The contents of MDA,MPO and TNF-α and W/D were lower in ISO group than in CLP group(P0.05 or P0.01).Conclusion In our experiment,intraperitoneal injection of ISO had anti-inflammatory and anti-oxidative effects,which provided partial protection from CLP-induced ALI in rats.
