几种糖对4℃贮存狂犬病病毒稳定性的影响 被引量:3

Effect of sugars on the stability of rabies virus stored at 4℃

摘要 为探讨糖类作为狂犬病液体疫苗保护剂的可行性,本研究将海藻糖、蔗糖和葡萄糖分别加入到狂犬病病毒(RV)培养物中,使终浓度为5%、10%和20%的悬液,以不加糖的RV培养物为对照,分别测定4℃贮存3、7、10、15、30d后的TCID50/mL和ELISA值。结果显示,TCID50/mL和ELISA值均随着存放时间的延长而逐渐降低;当含5%和10%海藻糖、5%蔗糖时,贮存3d后的TCID50/mL值比对照组高10倍以上,其中在含5%海藻糖时,TCID50/mL达10-8.5,表明RV滴度可以很好保持;而在加入葡萄糖各组的TCID50/mL和ELISA值均明显低于对照组。 Simple sugars have been used widely as lyophilized vaccine excipients,in order to investigate the potential of sugars as a stabilizer of liquid rabies viruses under 4℃,trehalose、sucrose and glucose were added as a final concentration at 5%,10%,20%,respectively,and taking no sugar adding rabies virus as control.TCID50/mL and ELISA values were detected under 4 ℃ after 3,7,10,15 and 30 d,respectively.The results showed that TCID50/mL and ELISA values decreased with extension of storage time;and on day 3,when the concentrations of trehalose at 5% and 10%,and sucrose at 5%,both TCID50/mL and ELISA values were higher than the control,and their TCID50/mL values were 10 times than the control,the TCID50/mL of 5% trehalose even reached up to 108.5,indicating that the virus titer can be maintained well under that;however,the titers in glucose contained groups were significantly lower than the control.
