藏红花对大鼠肝毒性的实验研究 被引量:14

摘要 目的研究藏红花是否具有肝毒性及引起药物性肝损伤的可能机制。方法大鼠随机分为5组,每组15只,标记为A、B、C、D、E组。用藏红花高、中、低浓度溶液(每毫升分别含生药80 mg、40 mg、20 mg)、0.9%氯化钠对A、B、C、D组大鼠灌胃,每日1次,每次1mL/100 g体重,6周;60%酒精对E组大鼠灌胃,每日2次,每次2 mL/100 g体重,6周。动态观察第2、6周末受试大鼠的一般情况、体重、肝功能指标,6周末肝组织光镜检查结果。结果A组大鼠血清碱性磷酸酶(ALP)水平随灌胃时间延长而升高,第2周、第6周分别为(190.5±37.8)U/mL、(253.5±46.6)U/mL,第6周ALP与对照组比较差异有显著意义(P<0.05)。肝组织病理:A组大鼠肝细胞排列紊乱,条索状结构消失,呈小泡状脂变,炎细胞浸润;B组肝细胞排列紊乱;C组无病理改变。结论高浓度藏红花具有肝毒性,线粒体失能可能为其发病机制。 Objective To study whether Saffron has liver toxicity,and the possible mechanism of liver injury.Methods Male Wistar rats were randomly divided into 5 groups,15 rats in each group.Rats in group A,B,C,D were fed with high,middle,low dosage Saffron liquid(80 mg/mL,40 mg/mL,20 mg/mL) respectively,0.9% NaCl 1 mL per 100 g once a day for 6 weeks.Rats in group E were fed with 60%alcohol 2 mL,per 100 g twice a day for 6 weeks.The rat's ordinary circumstances,weight and blood biochemistry were observed in 2th,6th week,and the microscopic examination was done in the end of the 6th week.Results The serum alkaline phosphatase activities went up along with the feeding time in high dosage group.The values were(190.5±37.8)U/mL,(253.5±46.6 U)/mL in 2th,6th weekend respectively.There was significant difference compared with group D in 6th week(P〈0.05).Microscope results showed that hepatocyte in group A swelled with the buildup of little fatty drop was in disorder,hepatocyte in group B was in disorder,hepatocyte in group C was normal.Conclution High dosage Saffron has hepatotoxicity.Mitochondria dysfunction might be possible mechanism.
