摘要
目的:观察表皮生长因子受体单克隆抗体mAb225对肿瘤细胞放射后凋亡的影响。方法:用流式细胞、双荧光染色、电镜3种方法分析单独应用mAb225或放射,以及两者联合应用所致肿瘤细胞的凋亡,并观察其时效和量效关系。结果:0.5μg/mL mAb225和6 Gy放射都能提高肿瘤细胞1倍左右的凋亡,两者联合可以提高5~6倍,在一定范围内,凋亡比例随作用时间和剂量的增加而增加。结论:mAb225可以明显提高肿瘤细胞的放射后凋亡。
Objective: To observe the effect of mAb225, a monoclonal antibody of epidermal growth factor receptor on tumor cell apoptosis after radiation. Methods: Tumor cell apoptosis treated with mAb225 or radiation were measured by FCM, bifluorescence staining and electron microscopy Results: Treatments of radiation alone (6 Gy) or mAb225 alone (0.5 μg/mL) procured apoptosis of SCC ceils by 2-fold, whereas combinations to both of them induced apoptosis to 5-6 fold. Conclusion: mAb225 could enhance apoptosis of tumor cell after radiation.
