用椭圆偏振术研究血清白蛋白和纤维蛋白原在硅片及聚氨酯薄膜表面的吸附行为 被引量:6

摘要 用椭圆偏振术研究了牛血清白蛋白(BSA)和人纤维蛋白原(FGN)在亲水硅片表面上的吸附行为。结果表明:蛋白质浓度大于5μg/ml时,相同蛋白质浓度下,BSA的最终吸附量小于FGN。在蛋白质浓度变化的情况下,BSA在吸附液蛋白质浓度约为10μg/ml时即达到饱和吸附(即最终吸附量不再随蛋白浓度提高而增加),而FGN达到饱和吸附时的吸附液浓度约为20μg/ml。BSA和FGN吸附液浓度同为30μg/ml时,BSA的吸附速率及饱和吸附量均大大低于FGN。对于同一蛋白质FGN,吸附液浓度30μg/ml时的吸附速率和饱和吸附量均明显大于浓度为7.5μg/ml时的吸附速率和饱和吸附量。用椭圆偏振术研究高分子材料表面蛋白质的吸附行为,主要问题是如何使生物材料表面达到椭圆偏振测量所要求的高光洁度和高反射率。本文发展了一种在抛光硅片基底上铺展高分子材料薄膜的方法,制备的样品符合椭圆偏振测量的要求。应用椭圆偏振术初步研究了FGN在聚氨酯表面的吸附行为,并与FGN在亲水硅片上的吸附行为进行了初步比较。结果表明,FGN在聚氨酯表面上的吸附速率和饱和吸附量均大大低于该蛋白质在亲水硅片表面上的吸附。提示亲水硅片的血液相容性比聚氨酯差得? dsorption behavior of bovine serum albumin(BSA) and human fibrinogen(FGN) was studies using ellipsometry. It is shown that the final amount of adsorbed BSA is less than that of FGN at equal protein concentration when the concentration is above 5μg/ml. With changed protein concentration, the adsorption of BSA showed to be saturated at concentration of 10μg/ml(namely, the final amount of adsorbed protein does not increase further with the increase of the protein concentration) while the concentration for FGN adsorption saturation is 20μg/ml. At equal protein concentration of 30μg/ml, both the adsorption speed and the final adsorbed amount of BSA were much less than those of FGN. For the same protein FGN, the adsorption speed and the final amount of adsorbed protein at protein concentration of 30μg/ml were obviously higher than those at concentration of 7.5μg/ml. The main problem for the ellipsometric study of protein adsorption at surfaces of polymeric materials is how to get surface smoothness and reflectivity high enough for the ellipsometric measurement. In this work, a method of polymer film spreading on surface of polished silicon chips has been developed. Samples prepared by this method accorded well with requirement of the ellipsometric measurement. Using ellipsometry, the adsorption behavior of FGN onto the polyurethane surface was preliminarily studied and compared with that on hydrophilic silicon surface. It is shown that the speed of FGN adsorption onto the polyurethane film and the final amount of the adsorbed protein are much less than those onto the hydrophilic silicon surface. It suggests that the hemocompatibility of hydrophilic silicon is much lower than that of polyurethane film.
