摘要
利用超声波对蜡样芽孢杆菌进行破碎处理,并测定蛋白质含量和SOD活性,探索以释放出更多高活性的SOD为目的的蜡样芽孢杆菌最佳细胞破碎条件及其对SOD活性的影响。研究结果表明,最佳的蜡样芽孢杆菌的超声波破碎条件为破碎浓度为10%、破碎功率为600W、破碎时间为5s、破碎次数为90次、变幅杆插入菌悬液深度为4cm。在此条件下菌体释放到破碎液内的蛋白质含量为0.3557mg/mL,而酶活力可达到493.38U/mg湿菌。
In the paper, protein concentration and SOD activity were determined after Bacillus cereus" were disrutped by ultrasonic wave in order to investigate the disruption condition for higher protein concentration and SOD activity released from disrupted cells. The results indicated that the primary condition for cell disruption was disruption time while protein contents reached 0.3557 mg/mLand SOD activity came to 493.38 U/mg wet cell under the optimized disruption condition of 10 % cell concentration at 600 W disruption power for 5 seconds repeated 90 times and ultrasonic probe inserted 4 cm deep into the cell liquid.
