摘要
用不同浓度的雌二醇或睾酮培养人肾小管上皮细胞株(HK-2)48h后,采用实时PCR检测人尿酸盐转运子mRNA的相对表达量。随雌二醇浓度的增加,人尿酸盐转运子mRNA表达水平明显增高(10^-12、10^-10、10^-8、10^-6mol/L时的相对表达量分别为1.03、1.05、1.67、1.87);随睾酮浓度的增加,人尿酸盐转运子mRNA表达水平明显降低(10^-7、10^-6、10^-5、10^-4mol/L时的相对表达量分别为0.56、0.34、0.25、0.19);培养条件下肾小管上皮细胞人尿酸盐转运子基因表达改变与性激素浓度的变化有关。
Human proximal tubule epithelial cell line, HK-2 cells, were cultured with various concentrations of estradiol or testosterone for 48 h. Human urate transporter (hUAT) mRNA was detected by realtime quantitative PCR. HUAT mRNA levels were up-regulated by estradiol ( 10^-12, 10^-10, 10^-8, 10^- 6 mol/L) in a dose-dependent manner ( relative expression median were 1.03, 1.05, 1.67, and 1.87), while down-regulated by testosterone. The relative expression median in 10^-7 , 10^-6 , 10^-5 , 10^-4 mol/L testosterone were 0.56, 0. 34, 0.25, and 0.19. In vitro, the expression of hUAT mRNA in HK-2 cells is associated with the concentration of sexual hormones.
