摘要
目的观察血管紧张素Ⅱ(AngⅡ)对正常血压和高血压患者的肠系膜动脉血管平滑肌细胞(VSMC)增殖及细胞内NADPH氧化酶-活性氧信号传导通路的影响。方法选取开腹手术正常血压和高血压患者肠系膜动脉培养的二至四代人VSMC作为标本,以四甲基偶氮唑盐(MTT)法、流式细胞仪、逆转录聚合酶链式反应(RT-PCR)分别测定细胞增殖率、细胞内活性氧及p22phox蛋白mRNA的表达。结果①与空白对照组比较,孵育组的细胞增殖率、细胞内活性氧物质生成量、细胞内p22phoxmRNA的表达均明显增高(均P<0.01)。②与正常血压组比较,给予AngⅡ刺激后,高血压患者的肠系膜动脉VSMC增殖率和细胞内p22phoxmRNA的表达量均明显增高(均P<0.01)。结论AngⅡ对高血压患者肠系膜动脉VSMC促细胞增殖作用较正常血压者更强。
Objective To investigate the effect of angiotensin Ⅱ on proliferation and redox signalling involving NADPH oxidase-derived reactive oxygen in cultured mesenteric artery smooth muscle cells from hypertensive patients.Methods Smooth muscle cells were isolated and cultured from mesenteric artery of Non-hypertension and hypertension patients.The proliferation activity of mesenteric artery smooth muscle cells was determined by MTT assay;reactive oxygen species was detected by flow cytometry;NADPH oxidase subunit p22^phox mRNA expression was detected by reverse transcription-polymerase chain reaction(RT-PCR).Results ①Compared with those of the control group,the proliferation activity,reactive oxygen species generation and p22phox mRNA expression in angiotensin Ⅱ stimulated cells were significantly increased(all P〈0.01).②Compared with cells derived from the non-hypertensive group,the proliferation rate of cells and p22phox mRNA expression of vsmc derived from the hypertensive group were significantly increased(all P〈0.01) in response to angiotensin Ⅱ stimulation.Conclusion Angiotensin Ⅱ has stronger effect on cellar proliferation of mesenteric artery smooth muscle cells in hypertensive patients.
