摘要
[目的]为合理开发利用紫锥菊产品提供理论依据。[方法]以早期妊娠的荷斯坦奶牛为试验动物,采用ELISA方法检测不同浓度狭叶紫锥菊提取物(PolinaceaTM)(6.3、20.0、60.0、180.0μg/ml)体外结合刀豆蛋白A(ConA,1μg/ml)和美洲商陆有丝分裂原(PWM,0.6μg/ml)对其外周血单核细胞(PBMC)增殖的影响。[结果]不同浓度PolinaceaTM均能促进正常奶牛PBMC增殖,180.0、6.3μg/ml PolinaceaTM刺激PBMC增殖的结果分别较对照高了近10和2倍;不同浓度PolinaceaTM结合ConA均可抑制PBMC增殖,其中180.0μg/ml PolinaceaTM结合ConA对PBMC增殖具有显著抑制作用;20.0、60.0μg/ml PolinaceaTM结合PWM可显著刺激PBMC增殖。[结论]紫锥菊提取物对奶牛外周血单核细胞增殖的影响具有剂量依赖性。
[Objective] The aim was to provide the theoretical basis for the rational development and utilization of Echinacea product.[Method]With early gravid holstein cows as the tested animals,the effects of Echinacea extract (Polinacea^TM) with different concn.(6.3,20.0,60.0,180.0 μg/ml) combined with concanavalin A(ConA ,1 μg/ml) and pokeweed mitogen (PWM,0.6 μg/ml) in vitro on proliferation of peripheral blood mononuclear cells(PBMC) of cows were studied with ELISA.[Result] Polinacea^TM with different concn.all could promote the proliferation of PBMC of normal cows,the proliferation results of PBMC stimulated by 180.0,6.3 μg/ml Polinacea^TM were higher 10 and 2 times than that of CK resp.Polinacea^TM with different concn.combined with ConA could inhibit the proliferation of PBMC,and 180.0 μg/ml Polinacea^TM combined with ConA had significantly inhibition on proliferation of PBMC.Polinacea^TM with 20.0,60.0 μg/ml combined with PWM could significantly stimulate the proliferation of PBMC.[Conclusion] The effect of Polinacea^TM on proliferation of peripheral blood mononuclear cells of cows had dose-dependent manner.
