摘要
目的探讨HMGN2对大肠埃希氏菌黏附Hela细胞的影响。方法采用Hela细胞模型,光镜以及扫描电镜观察大肠埃希氏菌ATCC25922黏附Hela细胞模型的建立。用以下三组不同方式,观察梯度抑菌浓度下(分别为5、10、15μg/ml)HMGN2对细菌黏附的影响。黏附抑制组:HMGN2、细菌、细胞共同孵育1小时;干扰模型组:细菌、细胞孵育1小时后,PBS洗去未黏附细菌,再加入HMGN2孵育2小时;细菌预处理组:HMGN2、细菌孵育2小时,离心洗去HMGN2,收集细菌,所得细菌再与细胞孵育1小时。用平板菌落计数法观测细胞内、外活菌数,并计算黏附指数以及黏附抑制率。结果光镜及扫描电镜可见细菌黏附于细胞表面,黏附模型构建成功。三组的结果均提示:在HMGN2梯度抑菌浓度下,细菌的黏附指数随着HMGN2的浓度的升高而减小;黏附抑制率则随着其浓度的升高而逐渐增大(P<0.05)。结论抑菌浓度下的HMGN2:对大肠埃希氏菌ATCC25922黏附Hela细胞有明显的抑制作用;可减低已经黏附于细胞表面的细菌的黏附能力;可能直接作用于细菌,导致其黏附能力降低。
Objective To investigate the effect of HMGN2 on E. Coli adhesion to Hela cell. Method Hela cell was used as an in vitro model for epithelium. The adhesion of ATCC25922 to Hela cell was observed by ordinary light mi- croscope and SEM. The effect of HMGN2 on bacteria adhesion was detected with concentration including 5, 10, 15μg/ ml. The study included 3 groups, including Adhesion inhibition group incubating with HMGN2, bacteria and cell for 1h, Interfere pattern group incubating with bacteria in lh and then with HMGN2 for 2h and Bacteria pre-treatment group incubating with HMGN2 for 2h, centrifuging and removing HMGN2, collecting bacteria and then incubating with cells for lh. The inside and outside cell viable bacteria was observed by APC, and then calculated the adhesion index and adhesion inhibition rate. Results The results showed that the adhesion index was decreased following the increase of HMGN2 concentration and the adhesion inhibition rate was significantly enhanced following the increase of HMGN2 concentration (P〈0.05), under MIC of HMGN2. Conclusion Under the MIC of HMGN2, HMGN2 could restrain the attachment of Escherichia eoli potent to Hela cell, HMGN2 could reduce the adhesion of bacterial which that had already adhered to the cell surface and HMGN2 may act on bacterial plump and lead to a reduction of adhesion.
