普通枇杷种内和种间杂种苗的RAPD鉴定 被引量:19

摘要 枇杷同其他木本果树一样,遗传上是高度杂合的,因此,获得真杂种是进行杂交育种等遗传改良的重要基础工作。以我国的特早熟优质枇杷品种早钟6号作母本,分别以西班牙的3个大果型品种作父本进行种内杂交;并以栎叶枇杷为母本与普通枇杷的解放钟作父本进行远缘杂交,4个杂交组合(ZJ、ZU、ZP和Lj)共获得493株杂种苗。运用RAPD分子标记,对这些杂交苗进行早期杂种鉴定,根据有无父本的特征带或新带型来鉴定其是否杂交成功,4个组合从100条RAPD引物中共计筛选出9条具有父本特征带的引物,鉴定454株杂种苗为真杂种。试验获得的真杂种苗可为枇杷品种的选育提供重要资源。 Loquats, being similar to other woody fruit trees, are highly heterozygous in heredity, therefore obtaining the true hybrid is the important essential work for genetic improvement via cross breeding. 493 hybrid individuals were obtained from the three different hybridization combinations of the early ripen and high quality Zaozhong No. 6 as the female parent and three big-fruit type Spanish loquats as the male parents and the distant hybridization combination of the oakleaf loquat as the female parent and the Jiefangzhong as the male parent. These hybrid individuals were identified in their early development stage using RAPD molecular markers. The true hybrids were confirmed according to the father fingerprinting characteristic band or the new fingerprinting. The nine RAPD primers selected from 100 primers were used for the analysis of 493 hybrid individuals and 454 true hybrids were obtained. The true hybrids obtained in our study provided the important resources for the breeding of loquat cultivars.
