SALL4 RNA干扰对THP-1白血病细胞生物学行为的影响

摘要 目的了解婆罗双树样基因4(SALL4)RNA干扰(RNAi)对THP-1白血病细胞生物学行为的影响。方法构建针对SALL4的特异性短发夹RNA(shRNA)干扰载体,并转染THP-1细胞,MTT法检测空白对照细胞,干扰载体转染细胞和阴性载体转染细胞的细胞增殖,流式细胞仪检测细胞凋亡和细胞周期,比较各组细胞的变化与区别。结果SALL shRNA载体转染的THP-1细胞在增殖能力上相对空白对照细胞及阴性对照细胞有明显下降,细胞凋亡率相对对照组细胞有明显上升,有丝分裂M期细胞比例减少。结论SALL4 RNAi后的THP-1细胞肿瘤细胞特征明显被抑制,SALL4对维持THP-1细胞恶性生物学特征有重要作用,SALL4可能是白血病发生发展过程中发挥重要作用的一种癌基因。 Objective To investigate the effects of SALL4 RNA interference on the biological behavior of THP-1 cells.Methods A SALL4-specific shRNA vector was constructed and transfected into THP-1 cells.The cell proliferation was measure by MTT in blank control,SALL4 shRNA and negative control groups.Apoptosis and cell cycle were assayed by flow cytometry.Results The proliferation ability of THP-1 cells tansfected by SALL4 shRNA was significantly decreased as compared with control and negative control groups.Apoptosis rate in SALL4 shRNA group was increased,and proportion of cells in M stage was reduced as compared with control groups.Conclusion The tumor cell characteristic of THP-1 cells was inhibited by SALL4 RNA interference obviously.SALL4 may play an important role to maintain the tumor cell characteristic of THP-1 cells and be an oncogene of leukemia.
