摘要
目的观察内抑制素对人脑胶质瘤细胞增殖的影响。方法①细胞培养:体外培养人脑胶质瘤细胞并进行光镜观察;②抑制因子试验:设立对照组与内抑制素实验组,采用四甲基偶氮唑蓝(MTT)法来判定该抑制因子对人脑胶质瘤细胞有无抑制作用;③细胞内游离Ca2+浓度测定:将特异性Ca2+荧光指示剂-Fluo-3用来负载人脑胶质瘤细胞,应用激光共聚焦显微镜检测细胞内游离钙的浓度。结果对照组光密度(OD)值明显高于各实验组(P<0.05);内抑制素能明显增加细胞内游离Ca2+的浓度,并且随药物浓度的增加而增加。结论内抑制素能够抑制人脑胶质瘤细胞的增殖且呈剂量依赖性,同时也能够增加胶质瘤细胞内游离Ca2+的浓度。
Objective To observe the inhibitive effect of endostatin on human glioma cells. Methods The control group and experimental groups were established in this research. Human glioma cells were cultured and observed under the inverted microscope. Methyl thiafolyl tetragolium (MTF) method was used to determine whether endostatin had the inhibitory effect on human glioma ceils. The specific indicator of fluorescence Ca2 + , dye Fluo-3, was applied in human glioma cells and laser scanning confocal microscope was used to measure the intracellular free Ca2+ concentration. Results The optical density (OD) value of control group was significantly higher than that of experimental groups (P 〈 0. 05). Endostatin could increase the intracellular free Ca2 + concentration with the increase of drug concentration. Conclusion Endostatin can inhibit the proliferation of human glioma cells in a dose-dependent manner, and it also can increase the intracellular free Ca2+ concentration.
