摘要
为探讨硫酸软骨素(CS)对深低温保存角膜内皮细胞的保护作用及机理,将牛角膜分成对照组及实验组Ⅰ~Ⅴ,每组6只角膜。对照组用75%二甲基亚砜(DMSO)为抗冻剂,实验组Ⅰ~Ⅳ除用75%DMSO外,还分别加用05%、25%、5%及75%CS,实验组Ⅴ抗冻剂仅有25%CS,没有DMSO。采用程序降温法深低温保存角膜,复温后经组织培养6~8小时检测内皮细胞密度及存活率,测量角膜厚度。结果:抗冻剂中含25%CS时,保存的角膜内皮细胞密度及存活率最高,且复温后角膜厚度恢复最快。但仅有25%CS,没有DMSO时,保存的角膜看不见内皮细胞结构。经含25%CS的抗冻剂保存的人角膜行穿透性角膜移植术后透明率高(5/6,8333%),但内皮细胞存活率(817%)较牛角膜内皮细胞存活率(944%)低。结论:CS能改善深低温保存的角膜内皮细胞活性。它虽不能代替DMSO,但可增强DMSO的抗冻效果。
To investigate the protective effects and their mechanism of chondroitin sulfate (CS) on corneal endothelial cells in corneal cryopreservation.Thirty six new born bovine corneas were divided into a control group and five experiment groups averasely (six corneas for each group).Cryoprotectant for control group was 7 5%dimethyl sulfoxide (DMSO),while cryoprotectants for experiment groups I to Ⅳwere respectively 0 5%,2 5%,5 0%,7 5%CS with 7 5%DMSO.Cryoprotectant for experiment group V was only 2 5%CS,without DMSO.Corneas were cryopreserved using program controlled Bio Freezer.Corneas were cultivated for 6~8 hours at 37℃ in an incubator equilibrated with humidified air containing 5%CO 2 after thawing,corneal endothelial cell density and survival rate were examined and stromal thickness was measured.Results Cryopreserved corneas had the highest endothelial cell density and survival rate when 2 5%CS was present in cryoprotectant with 7 5%DMSO.These corneas restored thickness the most quickly.However no endothelial cell structure were seen in corneas which were cryopreserved by only 2 5%CS without DMSO.Human cadaver corneas cryopreserved by cryoprotectant with 2 5%CS had high transparency rate after penetrating keratoplasty.But their endothelial cell survival rate was lower than that of bovine.Conclusions CS could improve viability of corneal endothelial cells in cryopreservation.It enhanced cryoprotective effects of DMSO on corneal endothelial cells,though it could not replace DMSO.
