摘要
以纯化的鸭乙型肝炎病毒表面抗原（DHBsAg）免疫BALB／C／c小鼠，取免疫鼠脾细胞与小鼠骨髓瘤SP2／O-Ag(14)融合，应用ELISA筛选出5B8和8G(11)2株抗体分泌滴度较高的杂交瘤细胞，二者所分泌的单抗仅与DHBaAg发生反应，而与其它9种禽类常见的病毒均不发生反应，其亚类鉴定分别属于IgM和IgG(2a)。用鸭乙型肝炎病毒（DHBV）阳性鸭血清中粗提的病毒悬液进行5B8株单抗介导的SPA胶体金试验，可见所有病毒颗粒均含有DHBsAg。用DHBs单抗夹心ELISA检测HBsAg、HBeAg、抗HBc阳性的病人血清，以及用HBV两对半酶标试剂盒检测DHBV阳性鸭血清，结果均为阴性，表明HBV与DHBV之间无抗原交叉性，而用这两山种方法检测家鹅血清中的乙肝病毒，发现家鹅体内的HBV与人HBV无抗原交叉性，但与DHBV呈现相同的表面抗原反应性。
Two hybridoma cell lines. designated as 5B8 and 8G(11) respectively. were derived by fusing SP2/0-Ag(14) myeloma cells and spleen cells of BALd/c mice immunized with purified duck hepatitis B virus surface antigen (DHBsAg). The monoclonal antibodies secreted by these two hybridoma cell lines respectively belonged to IgM and IgG(2a) isotype, as shown in ELISA, they reacted only with DHBsAg,but not with other viruses which infected fowls. In SPA-Golden Test mediated by 5B8 MAb, the golden particles were specifically linked to the surfaces of all kinds of DHBV particles concentrated from DHBV positive ducks, It Indicated that every kind of DHBV particle appeared in the sera of DHBV positive ducks contained DHBsAg. HBsAg, HBeAg and anti-HBc positive human sera, DHBV positive duck sera, domestic goose sera containing hepatitis virus particles were detected by Sandwich ELISA based on MAbs specific to DHBsAg and by ELISA with the kit of detection for HBV, all results showed that DHBV shared common antigen with hepatitis virus appeared in the sera of demestic geese, but DHBsAg didn't have cross-reaction with HBsAg.
