摘要
为探索ChMIL18-HN在毕赤酵母中最适宜的表达条件,采用小型摇瓶培养系统,通过对重组毕赤酵母菌X-33/pPICZαA-ChMIL18-HN在不同诱导时间、培养液pH值、甲醇诱导剂剂量及培养温度的条件下表达目的蛋白的分析,以对其表达条件进行优化。结果表明:重组菌X-33/pPICZαA-ChMIL18-HN在pH6.5,甲醇诱导浓度为1.5%,培养诱导温度为30℃的条件下诱导156h,目的蛋白的表达量最高。
To research the most optimization condition for expression of recombinant ChMIL18-HN in Pichia Pastoris,the P.pastoris X-33/pPICZαA-ChMIL18-HN was expressed in a small flask culture system.By studying the yield of target protein under different time of methanol inducing expression,different pH of medium,different inducing dose of methanol and different inducing temperature,the most appropriate expression conditions were determined.The results indicated that recombinant fusion protein was expressed to be the highest under pH6.5,methanol concentration 1.5%,culture-inducing temperature 30 ℃ for 156 hours.
