摘要
目的观察二氮嗪预处理对在体大鼠心肌缺血再灌注损伤(IRI)的保护作用。方法健康雄性SD大鼠36只,随机分成4组,每组9只。采用结扎左冠状动脉前降支(LAD)法制备心肌IRI模型。记录缺血再灌注前后不同时间点心功能指标,TTC染色测定心肌梗塞面积,检测血清CK-MB及LDH活性。结果结扎左冠状动脉前降支后:IRI组的CK-MB和LDH显著高于Sham组(P<0.05);IPC和DPC组较IRI组显著下降(P<0.05)。与IRI组比较,IPC组及DPC组梗死面积明显降低(P<0.05)。缺血30min、再灌注60min、120min时,IRI组、IPC组和DPC组与Sham组比较,±dp/dtmax、LVSP、LVEDP有显著的差异(P<0.05);IPC组和DPC组与IRI组比较,±dp/dtmax、LVSP差别有统计学意义(P<0.05)。结论二氮嗪预处理对在体大鼠缺血再灌注损伤心肌具有保护作用,可降低血清心肌酶含量、减少心肌梗死面积,维护心肌收缩和舒张功能。
Objective The study was aimed to investigate the protective effect of diazoxide preconditioning against myocardial IRI in rats in vivo.Methods 36 healthy male SD rats were randomly divided into 4 groups(n=9 each).Myocardial IRI injury model was prepared by ligating the LAD of rat.The index of cardiac function was recorded at the different time of before and after reperfusion.The infract size was measured by stained with TTC.The content of CK-MB and LDH in blood was Examined.Results After the LAD was ligatured,CK-MB and LDH of group IRI were higher than that of group Sham (P〈0.05),but in group IPC and DPC,CK-MB and LDH were low when compared with group IRI(P〈0.05).Compared with group IRI,infarct area size were obviously decreased (P〈0.05).After ischemic 30 min,reperfusion 60 min,reperfusion 120 min,there was obviously significant difference in±dp/dtmax、LVSP、LVEDP of group IRI,IPC and DPC when compared with group sham(P〈0.05);±dp/dtmax,LVSP of group IPC and DPC were higher than group IRI(P〈0.05).Conclusion There was protective effect of diazoxide preconditioning against myocardial IRI in rats in vivo,which can reduce levels of serum enzymes,reduce myocardial infarct size and maintenance myocardial systolic and diastolic function.
