大内皮素-1的定量ELISA法测定与临床应用 被引量:4

摘要 目的建立血浆大内皮素-1(BigET-1)的定量ELISA法测定,并探讨其应用价值。方法用定量ELISA法测定患者和健康者的血浆BigET-1。结果该法检出限为0.023pmol/L,线性为0.05-10.0pmol/L,平均回收率为99.1%,批内CV为3.10%,批间CV为5.23%。39例心绞痛和33例急性心肌梗死及52例原发性高血压患者的血浆BigET-1值分别为2.89±0.80,4.76±0.84和3.80±0.81pmol/L。结论该法灵敏、特异和准确,血浆BigET-1定量测定是诊断心血管疾病较好的实验指标。 Objective To establish enzyme-linked immunosorbent assay(ELISA) for the quantitative determination of big endothelin-1 (Big ET-1 in plasma and to investigate its clinical application. Methods Plasma Big ET-1 concentrations of patients and of normal controls were measured by ELISA. Results The limit of detection(LOD) was 0. 023 pmol/L. The linearity was 0.05-10.0 pmol/L. The average rate of recovery was 99.1%. Within-run CV was 3.10% and between-run CV was 5.26%. The plasma Big ET-1 concentrations of 39 patients with angina pectoris were 2.89+0. 80 pmol/L,of 33 patients with acute myocardial infarction were 4. 76± 0. 84 pmol/L and of 52 patients with primary hypertension were 3.80 ±0. 81 pmol/L. Conclusion This method is sensitive,specific and accurate. Quantitative detection of Big ET-1 is now better laboratory test for diagnosing cardiovascular diseases of patients.
