丹酚酸B对H_2O_2引起H9c2细胞氧化损伤的保护作用及可能靶点蛋白寻找 被引量:3

Protective Effect of Salvianolic Acid B on H9c2 Cells Injury Induced by H_2O_2 and Its Possible Target-related Proteins

摘要 观察丹酚酸B对H2O2引起H9c2细胞氧化损伤的影响,在分子水平上探讨丹酚酸B对H2O2诱导氧化损伤保护作用的可能机制。用H2O2刺激H9c2建立体外心肌细胞氧化损伤模型,MTT测定心肌细胞的存活率,Hochest33342染色观察细胞凋亡,并用蛋白质组学技术寻找丹酚酸B在心肌细胞中的可能蛋白质靶点。丹酚酸B可以提高心肌细胞在H2O2处理下的存活率,减少H2O2导致的细胞凋亡,丹酚酸B预处理组与单用H2O2组相比有8个差异表达蛋白,其中Heat shock 27 kua protein蛋白下调,cytochrome C oxidase、myo-sinlight polypeptide 6、myosin、myosin regulatory light chain 2、myosin regulatory light chain 9、tropomyosin和secreted acidic cysteine rich glycoprotein蛋白上调,差异表达的蛋白可能在丹酚酸B的保护机制中发挥重要作用。 The protective effect of Salvianolic acid B on the injury of H9c2 cells induced by H2O2 was checked and the possible target-related proteins of salvianolic acid B were studied using a proteomic method. After pre-treating with 10-8mol/L salvianolic acid B for 1 h (it is not treated in normal control group), H9c2 cells were treated with 400 μmol/L H202 for 3 h. The cell viability was measured by MTT assay and the apoptosis of cells was observed by Hochest33342 staining assay. Salvianolic acid B pretreatment could increase the cell viability and attenuate apoptosis of H9c2 cells induced by H2O2. In Proteomic study, the protein expression profiles were checked using two-dimensional electrophoresis. By comparing the protein expression profiles of salvianolic acid B-pretreated group and H2O2-treated group, 8 possible target related proteins of salvianolic acid B group were found. Among the 8 proteins, heat shock 27k protein was down-regulated whereas cytochrome C oxidase, myosin light polypeptide 6, myosin, myosin regulatory light chain 2, myosin regulatory light chain 9, tropomyosin and secreted acidic cysteine rich glycoprotein were up-regulated by salvianolic acid B pretreatment. These possible target-related proteins might play important roles in the protective effect of salvianolic acid B on H9C2 cells against H2O2-induced injury.
