摘要
为了解骨形态发生蛋白对关节软骨细胞的作用特点及意义,本研究用部分纯化的牛骨形态发生蛋白(bBMP)对胎儿关节软骨细胞进行诱导,并用人Ⅰ、Ⅱ、Ⅲ、Ⅳ型胶原的前胶原cDNA探针对被诱导关节软骨细胞进行原位杂交。结果发现,bBMP可明显增加被诱导软骨细胞中Ⅰ型胶原的表达,中断Ⅱ型胶原的表达,并轻度增加软骨细胞中Ⅲ型胶原的表达,但对Ⅳ型胶原的表达影响不大。本研究认为,bBMP诱导的软骨细胞胶原基因表达的这种改变是软骨细胞在向成骨细胞样细胞方向分化的标志。
o understand the characteristic and significance of action of bone morphogenetic protein on articular chondrocytes,partialy purified bovine bone morphogenetic protein (bBMP) was used to induce embryo articular chondrocytes; the induced articular chondrocytes were hybridized in situ with cDNA probes of human procollagens of Ⅰ、Ⅱ、Ⅲ、Ⅳ types. The results showed that bBMP could increase the expression of type Ⅰ collagen of the induced articular chondrocytes significantly, stopped their type Ⅱ collagens expression and slightly increased the expression of type Ⅲcollagen , but the expression of type Ⅳ collagen was not seriously influenced. It was believed that the change of the expression of collagen genes was the mark of chondrocytes differentiating towards osteoblastlike cells.
