甲型肝炎病毒H2株单克隆抗体的制备及鉴定

摘要 目的为甲型肝炎病毒(HAV)的诊断和相关研究提供高效价的抗体来源。方法用细胞培养获得的H2株HAV免疫BALB/C小鼠,经细胞融合技术,ELISA筛选和有限稀释克隆化培养。结果获得3株能稳定分泌抗HAV单克隆抗体(McAb)的杂交瘤细胞株,命名为6-A8、6-F2和1-H3。经鉴定,其上清和腹水的抗体效价分别为1:(2048~16824)和1:(65536~524288)。6-A8和6-F2属IgG2a亚型,1—H3属IgGl亚型。染色体数分布在92~98之间。3株细胞株McAb分别能与HAV不同抗原位点结合。结论3株能稳定分泌抗HAVMcAb的杂交瘤细胞株成功建立,并获得高效价的抗HAVMcAb。 Objective To obtain monoclonal antibody against hepatitis A virus. Methods BALB/C mice were immunized with the purified HAV HE strain from cell culture. The lymphocytes from the splenic tissues of the mice were fused with SP2/0 myeloma cell screening with ELISA, limiting dilution technique and expanded into larger cell populations. Results Three lines of hybridoma cell secreting McAb against HAV were expanded. By evaluating, two strains were IgG2a subset, and the other one was IgG1 subset. The fiter of McAb in supematant of culture and ascites fluids were 1:(2048-16 824) and 1 : (65 536-524 288)respectively. The chromosome number was 92-98. These McAb could recognized different epitupes of HAV-antigen. Conclusions The three lines of hybridoma cell secreting McAb against HAV are successfully established, and also obtained high rites of McAb.
