去甲斑蝥素对CXCR4表达及MCF-7细胞迁移的影响 被引量:3

摘要 目的探讨去甲斑蝥素(NCTD)抑制乳腺癌细胞MCF-7侵袭转移的机制。方法体外培养人乳腺癌MCF-7细胞株,采用MTT法检测NCTD对MCF-7细胞增殖抑制浓度;建立体外细胞趋化转移模型分析200,400,600μmol/LNCTD抑制MCF-7细胞迁移的能力;realtime PCR、Western blot分析不同浓度药物处理后的MCF-7细胞中CXCR4及CXCL12表达。结果NCTD可抑制MCF-7细胞增殖,具有明显的量效关系,24h的IC50为582μmol/L;在体外条件下,NCTD可以抑制CXCL12所趋化的细胞迁移;同时可以抑制MCF-7细胞中的CXCR4表达。结论NCTD通过下调CXCR4的表达抑制乳腺癌细胞MCF-7的迁移。 Purpose To investigate the effect and mechanism of norcantharidin(NCTD) on invasion of human breast cancer cell line MCF-7 in vitro.Methods MTT assay was used to determine MCF-7 cell proliferation,Transwell kit was used to determine cell migration,treated with NCTD on 200,400,600 μmol/L,to detect the expression of CXCR4 and CXCL12 in the control and treatment groups by real time PCR and Western blot.Results NCTD had inhibitive effects on proliferation and invasion of human breast cancer cell line MCF-7 in a dose-dependent manner,with the IC50 value of 582 μmol/L at 24 h,and the expression of CXCR4 was decreased in NCTD groups in vitro.Conclusion NCTD in vitro inhibits invasion and metastasis of human beast cancer cell line MCF-7 through the down regulation to the expression of CXCR4.
