流式细胞术检测非小细胞肺癌外周血循环内皮细胞方法初探 被引量:5

摘要 目的探索流式细胞术单种CD分子(CD146)标记法和三种CD分子(CD45CD31CD146)标记法对非小细胞肺癌外周血循环内皮细胞(circulating endothelial cells,CECs)检测效果的影响。方法采用流式细胞术单种CD146和三种CD分子标记法检测60例非小细胞肺癌患者及20例健康人外周血CECs。结果流式细胞术单种CD146和三种CD分子标记法对非小细胞肺癌患者和健康人外周血循环内皮细胞检测成功,两种方法显示肺癌患者外周血CECs均高于健康对照,差异有统计学意义(P<0.05)。两种方法测定CECs的结果呈正相关,有统计学意义(r=0.834,P<0.01)。精密度试验显示三种CD分子标记循环内皮细胞法优于单种CD146标记法。结论单种CD146和三种CD分子标记非小细胞肺癌患者和健康人外周血细胞均可经流式细胞术用于测定CECs,三种CD分子标记循环内皮细胞法优于单种CD146标记法。 Objective To explore the influence of circulating endothelial cells in non-small cell lung cancer(NSCLC) patients with single fluoresce-labeled monoclonal antibody(CD146) and three fluoresce-labeled monoclonal antibodies(CD45CD31CD146) by flow cytometry,and to compare the difference with healthy volunteers.Methods CECs were detected by flow cytometry with single and three fluoresce-labeled monoclonal antibodies in peripheral blood of sixty NSCLC patients and twenty healthy volunteers.Results CECs were detected successfully by flow cytometry with single fluoresce-labeled monoclonal antibody and three fluoresce-labeled monoclonal antibodies,there were statistical significances in two methods between patients and healthy volunteers(P〈0.05) respectively.There was significant positive correlation(r=0.834,P〈0.01) between the results of two methods.The single fluoresce-labeled monoclonal antibody(CD146) of flow cytometry was better than three fluoresce-labeled monoclonal antibodies(CD45CD31CD146).Conclusion Two methods can be used in detecting CECs by flow cytometry,the efficiency of single fluoresce-labeled monoclonal antibody by flow cytometry was superior to three fluoresce-labeled monoclonal antibodies in detecting circulating endothelial cells.
