摘要
哮喘易感基因的研究正在世界各地许多实验室蓬勃开展,由于此种疾病的复杂性和异质性等原因,哮喘易感基因的鉴定依然任重而道远。目前的研究表明,哮喘所涉及的染色体达10多条,与其关联或连锁的候选易感基因多达170多个,但多数研究结果的一致性和重复性较差。分析其中的原因,除了哮喘的复杂性、种族的差异、病例的选择与分层的不同外,研究策略、技术路线和方法应该是导致结果不一致的主要原因之一。本文基于系统生物学理论与候选基因的方法,系统阐述一种哮喘易感基因研究的新策略。
Efforts to identify the asthma s usceptible genes have been engaged in many laboratories around the world. Because of the complexity and heterogeneity of the disease it is till a daunting task. Although the present results have been demonstrated that more than 10 chromosomes and 170 genes associated or in linkage with asthma and asthma related phenotypes, most of the reports on asthma susceptible genes are either inconsistency or controversial. Many factors, including the inherent complexity of asthma, ethnic diversity, sample size, inadequate control, and asthma stratification, can explain these inconsistencies, which the design and methodological issues also are main factors. In this review, we propose a new strategy on study of susceptibility genes of asthma based on theory of systems biology and candidate gene approach.
