摘要
目的建立一种新的复合式阿尔茨海默病(AD)大鼠模型,用于AD及其药物治疗的研究。方法SD雄性大鼠随机分为正常组、假手术组、模型组。在大鼠左侧海马区内注β-淀粉样蛋白1-42肽段(Aβ1-42)和鹅膏蕈氨酸(IBO)混合液造成AD模型,通过水迷宫试验比较各组差异。结果在定位航行中,模型组潜伏期明显比正常组和假手术组延长(P<0.01)。结论Aβ1-42合并IBO诱导阿尔茨海默病大鼠模型在一定程度上较好的模拟AD的发病特点,可作为AD及其药物研究的一种新模型。
Objective To establish a novel complex rat model with Alzheimer's disease fAD) for research of AD and its treatment. Methods The Sprague - Dawley male rats were randomly divided into normal group, sham - operated group, and model group. The Sprague- Dawley rat models with AD were induced by injection of beta- amyloidl -42(A131 -42)and ibotenic acid(IBO)into left hip- pocampus. All rats were evaluated hy Morris water maze test. Results The latency of place navigation in model group was significant- ly longer than that in normal group and sham - operated group (P〈0.01). Conclusion There were some characteristics in rat model with AD induced by injection of Aβ1-42 and IBO into the area of left hippocampus similar to human AD. It might be used as a novel model to study AD and its drug treatment.
