摘要
目的:建立一个以最低保护力的抗体水平(15IU/ml)为临界值的风疹病毒特异性IgG抗体的检测方法。方法:用优选的抗原和具有变色功能的标本稀释液,用世界卫生组织提供的标准品标定标准曲线,以常规间接ELISA方法进行检测。结果:对检测方法的各个环节进行了比较研究,确定了检测方法的最佳工艺流程。对1005例育龄妇女血清标本进行检测,并与意大利Sorin公司的试剂检测结果进行比较,两种方法呈高度相关(Kappa值为0.85)。结论:本文建立的方法具有敏感、特异、简便、快速、可重复性好等特点,适用于育龄妇女风疹病毒抗体水平的筛查。
Objective:To establish a method for detecting rubella virus(RV)specific immunoglobulin G(IgG)antibodies using 15IU/ml of anti-RV-IgG defined as minimum immunity criteria of preventable RV infection.Methods:The measurement of anti-RV-IgG was conducted by indirect enzyme-linked immunosorbent assay(ELISA)using optimal RV-antigen and the sample dilution buffer with a color change function.Standard curve were calibrated with international standard for anti-rubella immunoglobulin substance provided by World Health Organization.Results:A technical research on each step of the method for detecting RV-IgG was performed.And the optimal technique flowchart was defined.The result of this method was compared with that of the assay using a Sorin kit by measuring anti-RV-IgG in the serum samples of 1 005 women of reproductive age,showing a high consistency(Kappa value:0.85).Conclusion:The specific,sensitive and reliable method we established could be used for RV antibodies screening among women of reproductive age.
