摘要
目的针对目前没有适合中生菌素产生菌的合成培养基的现状,进行中生菌素产生菌发酵合成培养基的设计,并对其进行优化,以期为以后中生菌素产生菌发酵生产、营养生长、代谢、遗传育种和产素机制的研究提供基础。方法顺序通过无机氮源和有机氮源的筛选、正交试验,并利用统计学软件SPSS V13.0对实验数据进行分析,确定并优化中生菌素产生菌发酵合成培养基的组成成分。结果最终确定了中生菌素产生菌发酵合成培养基,该培养基在定量加入微量元素的基础上,组成成分为谷氨酸钠0.5%,葡萄糖1.5%,可溶性淀粉1.5%,NH4Cl 0.3%,KH2PO4 0.02%,MgSO4 0.025%,NaCl 0.5%,CaCO3 0.3%。结论经过发酵验证,该培养基的产素能力可达到2000μg/mL左右,虽然比天然成分培养基低,但可以满足以后营养生长、代谢、遗传育种和产素机制等的研究。
t Objective To design a chemically defined medium for Streptomyces lavendulae var. hainanensisn which produce zhongshengmycin. Methods The inorganic and organic nitrogen sources used in the chemical defined medium were screened. Then the medium was optimized with an orthogonal experiment. The experimental data were further analyzed with statistic software SPSS V13.0. Results The composition of the chemically defined medium was decided through these experiments. It contains 0.5% sodium glutamate, 1.5% glucose, 1.5% soluble starch, 0.02% NH4C1, 0.02% KH2PO4, 0.025% MgSO4, 0.5% NaCI, 0.3% CaCO3 and lmL trace elements buffers. Conclusion The zhongshengmycin production level reached 2000μg/mL in the chemically defined medium, which has great potentiality for the future research.
