摘要
目的检测人正常胃黏膜及胃癌组织中gp78和KAI1表达水平并探讨两者在胃癌发生过程中的临床意义。方法分别应用Westernblot和免疫组织化学方法检测18例正常胃黏膜和65例胃癌组织中gp78和KAI1的表达情况。结果Westernblot结果表明,65例胃癌组织中有25例(38.5%)gp78水平升高,27例(41.5%)KAI1水平降低,其中13例(20.0%)既出现gp78水平升高又出现KAI1水平降低。与正常胃黏膜组织比较,胃癌组织中gp78的表达水平明显升高(P<0.01),而KAI1的表达水平明显下调(P<0.01)。免疫组织化学提示,65例胃癌组织中21例(32.3%)gp78阳性表达率升高,22例(33.8%)KAI1阳性表达率降低,其中10例(15.4%)既出现gp78阳性表达率升高又出现KAI1阳性表达率降低。Spearman相关性分析表明,胃癌组织中gp78和KAI1表达水平呈现明显的负相关(r=-0.908,P<0.05)。同时,随着淋巴转移、肿瘤浸润以及脉管浸润程度加重,胃癌组织中gp78表达水平明显升高,而KAI1表达水平显著降低。而在年龄、性别等因素上gp78和KAI1表达水平差异无统计学意义(P>0.05)。结论gp78和KAI1在胃癌的发生、发展过程中起着重要的作用。gp78的过表达导致抑癌基因KAI1的降解增多可能是胃癌浸润、转移的机制之一。
Objective To determine the expression levels of gp78 and KAI1 in human gastric carcinoma tissue and investigate their clinical significance in the development of gastric carcinoma.MethodsWestern blot analysis and immunohistochemical staining were employed to assay gp78 and KAI1 expressions in 18 samples of normal gastric mucosa and 65 samples of gastric carcinoma tissue.ResultsWestern blotting results indicated that gp78 protein level was increased in 25 cases(38.5%)and KAI1 protein level was decreased in 27 cases(41.5%).Meanwhile,simultaneous increased gp78 and decreased KAI1 levels were detected in 13 cases(20.0%).Immunohistochemical results showed that gp78 level was up-regulated in 21 cases(32.3%),and KAI1 level was down-regulated in 22 cases(33.8%).Further,simultaneous increased gp78 and decreased KAI1 levels were found in 10(15.4%)out of 65 cases.Spearman correlation assay suggested there was a significant negative correlation between gp78 and KAI1 expression(r=-0.908,P〈0.05).Respectively,the expression level of gp78 and KAI1 was significantly enhanced and reduced along with the aggravation of lymphatic metastasis,tumor infiltration,and vessel infiltration.However,no statistical difference was seen in age and sex between groups(P〉0.05).ConclusionGp78 and KAI1 play important roles in the genesis and development of gastric carcinoma.Compared with the normal gastric mucosa tissue,the expressions of gp78 and KAI1 are remarkably up-regulated and down-regulated,respectively.In fact,overexpression of gp78 results in more degradation of KAI1,which might be responsible for infiltration and metastasis of gastric carcinoma.
