摘要
利用复合酶在其最优条件下降解高温蒸煮玉米秸秆,确定复合酶降解最优条件:0.8g底物(蒸煮秸秆0.6144g)加入30mlpH4.8柠檬酸缓冲液,调pH4.8,121℃灭菌20min,待冷却,无菌操作下加入20mg纤维素酶、10mg木聚糖酶、6mgβ–葡聚糖酶、1.3mg果胶酶,在50℃、100r/min条件下酶解16h,降解率为70.03%。此法降解率已非常接近玉米秸秆理论降解率(约74%),对玉米秸秆降解研究具有一定参考价值。
Steam-pretreated corn stover was degraded by complex enzyme under the optimum conditions. The optimal conditions are: 0.8g substrate ( Steam-pretreated corn stover 0.6144g ) is put in 30 ml citrate buffer solution ( pH4.8 ) and adjusted pH4.8 before pasteurization at 121℃ for 20 min. Then put 20 mg cellulase, 10 mg xylanase,6 mg β-glucanase and 1.3 mg pectase in it, after cooling down. Finally, enzymatic degradation rate is 70.03% under the conditions of rotate speed 100 r/min, temperature 50℃ and time 16 h. The degradation rate of the method is very close to the theoretic degradation rate about 74%. So this article has a very high referential value for research on degradation of corn stover.
