褶皱臂尾轮虫对2种细菌的摄食及其生长情况研究

摘要 [目的]研究活饵料对抗原物质的吞噬及存留情况。[方法]以不同浓度的嗜水气单胞菌(活菌)和迟缓爱德华氏菌(灭活细菌)作为饵料定时投喂人工孵化的褶皱臂尾轮虫,破碎轮虫后统计其体内的细菌数;取染色后的细菌投喂轮虫,显微观察轮虫消化道内细菌的存留时间。[结果]6d后活菌组轮虫密度最大为70.3个/ml,灭活组轮虫密度为13.8个/ml;活菌组轮虫的平均抱卵率分别为28.80%、38.02%、33.91%、21.40%、18.78%、15.50%,灭活组轮虫的平均抱卵率分别为56.39%、61.66%、53.54%;轮虫对细菌的消化时间与所处温度呈负相关,但温度过高也会导致消化时间延长;每个轮虫摄食嗜水气单胞菌26785.3cfu,摄食迟缓爱德华氏菌3652.2个。[结论]活体生物可对抗原物质进行有效包囊。 [Objective] The aim was to study the phagocytosis and retention condition of live bait on antigens.[Methods] The Aeromonas hydrophila (live bacteria) and the Edwardsiella tarda (inactivated bacteria) with different concn.were taken to feed the artificial incubated Brachionus plicatilis termly.The number of bacteria in the body of B.plicatilis was calculated after breaking of the B.plicatilis.The stained bacteria were taken to feed the B.plicatilis and the retention time of the bacteria in digestive tract of the B.plicatilis was observed with microscope.[Results] After 6 d,the maximal density of B.plicatilis in live bacteria group reached 70.3 ind/ml,and that in inactivated bacteria group was 13.8 ind./ml.The average spawning rates of B.plicatilis in live bacteria group were 28.80%,38.02%,33.91%,21.40%,18.78% and 15.50% resp.and those of B.plicatilis in inactivated bacteria group were 56.39%,61.66% and 53.54% resp..The digestive time of B.plicatilis on bacteria had negative correlation with the temperature,but the excessively high temperature could delay the digestive time.Each B.plicatilis could intake 2 6785.3 cfu A.hydrophilas and 3 652.2 cfu E.tarda.[Conclusion] The antigens could be effectively inwraped by live organisms.
