摘要
目的观察表没食子儿茶素没食子酸酯(-)(EGCG)对昆明(kunming,KM)小鼠早胚体外发育的影响,为改善小鼠早胚体外培养体系奠定实验基础。方法以空白M16培养液为对照组,M16中添加浓度为0.1μg/mL、1μg/mL、10μg/mL、20μg/mL EGCG为实验组,收集KM小鼠1-细胞胚进行体外连续培养,计数各组发育至2-、4-细胞胚、桑椹胚和囊胚等各个阶段的数目,并以2-细胞胚为基数,计算各组至不同阶段的发育率。结果添加EGCG实验组发育到桑椹胚和囊胚的比率明显高于对照组,其中以添加浓度为10μg/mL和20μg/mL的EGCG实验组的效果最显著,其桑椹胚发育率分别为77.3%和78.7%,而对照组只有43.2%(P<0.01)。10μg/mL和20μg/mL的EGCG实验组的囊胚发育率分别为53.0%和47.3%,也明显高于对照组(19.2%,P<0.01)。结论EGCG可以显著提高KM小鼠1-细胞胚体外发育到桑椹胚及囊胚的比率,以10μg/mL和20μg/mL浓度的EGCG效果最显著。
Objective To investigate the effect of(-)epigallocatechin gallate(EGCG)on the development of Kunming(KM)mouse early embryos in vitro.Methods The KM 1-cell embryos were collected and cultured in M16 medium supplemented with EGCG(0.1,1,10,20μg/mL)or without EGCG(control).The number of KM 1-cell embryos developed to the 2-cell embryos,4-cell embryos,morula and blastocysts was counted under an inverted microscope.Results The M16 media containing EGCG had higher developmental rates from 2-cell embryos to 4-cell embryos,to morula and blastocysts than the control.Addition of 10μg/mL and 20μg/mL of EGCG significantly increased the developmental rates of morulae and blastocysts compared with the control(P〈0.01).Conclusion EGCG enhances the development of KM 1-cell embryos to morula and blastocysts.
