浙江沿海地区汉族人A型及AB型血型的基因亚型研究 被引量:5

摘要 目的:探讨浙江沿海地区汉族人A型及AB型血型的基因亚型分布情况,为输血及血库血量储存提供理论依据。方法:采用序列特异引物聚合酶链反应(PCR-SSP)检测该地区汉族成人体检者血样本DNA基因亚型,A型280份,AB型110份,根据PCR-SSP反应产生的特异性阳性条带判定相应的基因亚型。结果:A型样本分型成功278份,AB型样本分型成功107份。A型血基因亚型以A/A201、A/O及A201/O较多,占97.84%,而A201/A201及A/A占2.16%;AB型血中基因亚型A/B型(51.40%)及A201/B型(48.60%)所占比例较为接近。A型及AB型血型的基因亚型分布在男女性别之间无显著性差异(P>0.05)。结论:浙江沿海地区汉族人群A型血中存在多种基因亚型,A型血型遗传多态性以及不同亚型之间的抗原性差异需进一步研究。 Objective:To investigate the genotypes of the ABO blood group in the Han nationality of littoral in Zhejiang Province.Methods:Three hundred and ninety blood samples from unrelated Han nationality of littoral in Zhejiang Province including 280 case of blood group A and 110 case of blood group AB,were genotyped by the polymerase chain reaction with sequence-specific primers(PCR-SSP).Genomics DNA was extracted and amplified by four sequence specific primers,and the typing results were identified corresponding to the special positive strip of PCR reaction.Results:278 blood group A samples and 107 blood group AB samples were genotyped successfully.In group A,genotypes of A/A201,A/O,A201/O accounted for 97.84% of total,while A201/A201 and A/A accounted for 2.16%.In group AB,the frequencies of gene subtype A/B and A201/B were 51.40% and 48.6% respectively.There are no significant difference with blood group A and blood group AB of men and women.Conclusion:The distribution of genotype frequencies of Han nationality of littoral in Zhejiang Province is significantly different with that of other populations.
