Proliferation and Osteogenic Differentiation of Bone Mesenchymal Stem Cells Induced with bFGF Combined Dexamethasome

摘要 目的:研究碱性成纤维细胞生长因子(bFGF)和地塞米松联合对骨髓间充质干细胞(BMSCs)增殖和成骨分化的影响。方法:以第三代BMSCs为实验材料,分别给予10%FBS的DMEM培养基,含80μg/LbFGF的10%FBS的DMEM培养基,含bFGF80μg/L+地塞米松10-8mol/L+β-甘油磷酸钠10mmol/L+维生素C50mg/L的10%FBS的DMEM培养基处理,通过计数BMSCs细胞数评价其增殖能力,同时行碱性磷酸酶(ALP)活性检测及茜红素染色评价其成骨分化的能力。结果:与对照组相比较,bFGF可以显著提高骨髓间充质干细胞的增殖能力,bF-GF和地塞米松联合不仅促进了骨髓间质干细胞的增殖,而且促进碱性磷酸酶的表达和钙结节的形成。结论:bFGF和地塞米松联合促进了骨髓间充质干细胞的增殖和成骨分化。 Objective To investigate the effect of basic fibroblast growth factor(bFGF) combined dexamethasome on the proliferation and osteogenic differentiation of bone mesenchymal stem cells(BMSCs). Methods The third generation BMSCs were cultured with 10% FBS DMEM medium,10% FBS DMEM medium containing 80 p,g/L bFGF,10% FBS DMEM me- dium containing 80 I.Lg/L bFGF + 10 -Smol/L dexamethasome + 10 mmol/L ]3-sodium glycerophosphate + 50 mg/L vitamin C, respectively. The ability of proliferation was evaluated with cell count and the ability of osteogenie differentiation was e- valuated with the activity of alkaline phosphatase (ALP) and Alizarin red staining. Results Compared with control group, the proliferation ability of BMSCs could significantly promote with bFGF, however bFGF combined with dexamethasome could promote not only the proliferation ability,but also the expression of ALP and the formation of bone nodules. Conclusion bFGF combined dexamethasome could promote the proliferation and osteogenic differentiation of BMSCs.
