摘要
目的研究内皮素-1反义脱氧寡核苷酸(ET-1AS-ODN)对链脲佐菌素(streptozotocin,STZ)诱导的糖尿病肾病大鼠肾组织超微结构的保护作用及其机制。方法一次性腹腔注射STZ60mg/kg,建立大鼠糖尿病肾病模型,并观察ET-1AS-ODN预防性给药对大鼠肾组织超微结构的保护作用。结果实验动物饲养5周后,与生理盐水对照组相比,糖尿病肾病模型组大鼠一般状况差,体重明显降低(P<0.01),肾损伤明显;与DM组比较,ET-1AS-ODN各组大鼠精神状态有所改善、电镜下肾组织超微结构损伤减轻。结论内皮素-1反义寡核苷酸可减轻STZ诱导的糖尿病肾病大鼠的肾损伤,对肾组织超微结构具有保护作用。
Objective To investigate the Pprotective effects of endothelin-1 mRNA antisense oligodeoxynucleotide(ET-1AS-ODN)on the glomerulus ultrastructure in diabetic nephropathy(DN)and its mechanism in diabetic rat model.Methods Diabetic rats were induced by intraperitoneal administration of streptozotocin(STZ)at 60mg/kg.ET-1AS-ODN was administrated intravenously to the treatment group and its protective effects were observed.Results Compared with the control group,the general condition and body weight of the DN group decreased significantly(P0.01),while the renal indexes increased significantly.Proliferated mesangium,thinkened glumerular basement membrane and shortened podocytes were observed in DN group subjects.Compared with the DN group,the general condition and nephrological renal histological injury in the ET-1 AS-ODN treatment group subjects improved significantly.Conclusion Endothelin-1mRNA antisense oligodeoxynucleotide inducinged inhibition of endogenous ET-1 has a protective effect on the glomerularumerular ultrastructure and can prevent glomerular sclerosis in diabetic nephropathy rats.It indicates that ET-1 does play an important role in the pathogenesis diabetic nephropathy.
