Genetic monitoring of the three Inbred Strain Mouse

摘要 目的检测新培育的IRM-1近交系小鼠的基因纯合性,并与IRM-2、615近交系小鼠进行比较,确定其遗传特性。方法按照国家标准GB/T14927.1-2001、GB/T14927.2-2001遗传检测操作规程,对IRM-1近交系小鼠进行Akpl等25个遗传生化标记基因纯合度、组织相容性基因纯合度的测定,用同系异体间进行移植试验;以及毛色基因测试。结果IRM-1近交系小鼠的25个遗传生化标记基因均为纯合体,其中有5个生化标记基因多态性位点与IRM-2近交系小鼠不同,有8个生化标记基因多态性位点与615近交系小鼠不同;皮肤移植100d后,未见排斥现象;毛色基因测试,F1的毛色全部为野生色,基因型为aaBBCCDD。结论IRM-1小鼠遗传纯合度已达国家标准,是基因位点高度纯合的近交系小鼠。 Objective To examine the genetic homozygosity of the IRM - 1 mouse bred recently and compare with the IRM -2 and 615 mice to determine IRM - 1 mouse genetic characteristics. Methods According to the national stardard GB/T14927.1 -2001 and GB/T14927 2 -2001 genetic operating instructions, the Akpl ete 25 biochemical markers were detected to observe the genetic homozygosity of the IRM - 1 inbred mouse . In addition, the histoeompatibility genetic homozygous determination, transplantation experiment between homologous series variant, and hair color test by mating with BALB/e were all carried out. Results All the 25 genetic biochemical markers are homozygote, among which five biochemical markers gene polymorphism sites are different from the IRM -2 mouse, eight biochemical markers gene polymorphism sites are different from the 615 mouse. No dropping graft was found after transplanting100 days. The coat color genes analysis revealed that hair color of first generation hybrid was wild color, whose gene type was AAhbCCDD. Conclusions The IRM - 1 inbred strain mouse is high homozygositous , whose degree has reached international standard.
