摘要
目的探讨益母草碱(Leo)和水苏碱(Sta)合用抗小鼠急性心肌缺血的作用。方法采用皮下注射异丙肾上腺素(Iso)建立小鼠急性心肌缺血模型,观察Leo和Sta合用对小鼠心电图T波的影响,并且根据金氏公式计算q值,评价合用效应,测定小鼠血清中丙二醛(MDA)含量、乳酸脱氢酶(LDH)活力,HE染色观察心肌组织病理损伤情况。结果与模型组比较,合用组(Leo2.5mg/kg+Sta5mg/kg)、(Leo5mg/kg+Sta10mg/kg)显著抑制T波变化;降低血清中的MDA含量和LDH活力;同时改善异丙肾上腺素诱导的心肌缺血的病理损伤。应用金氏公式计算q值在0.85~2.0范围内,评价合用组抑制T波变化效应为单纯相加或增强作用;合用组(Leo5mg/kg+Sta10mg/kg)与单用Leo5mg/kg比较MDA含量降低明显,与单用Sta10mg/kg比较LDH活力降低明显;合用组(Leo2.5mg/kg+Sta5mg/kg)、(Leo5mg/kg+Sta10mg/kg)改善小鼠的心肌组织病理损伤优于单用Leo组和Sta组,仅见少量的心肌纤维收缩。结论Leo和Sta合用抗小鼠急性心肌缺血的作用优于单用。
Objective To explore the effects of the combination of Leonurine(Leo)and Stachydrine(Sta)on acute myocardial ischemic in mice.Methods The acute myocardial ischemic model was made by using subcutaneous injection of isoproterenol(Iso)in mice,T wave of electrocardiogram(ECG)was observed,and the activitise of lactate dehydrogenase(LDH)and the contents of malonaldehyde(MDA)in serum were measured,Jin's formula was applied to analyze the combination of Leo and Sta,The histopathological changes of myocardium were observed by HE staining.Results Both the combination Leo 2.5 mg/kg with Sta 5 mg/kg and Leo 5 mg/kg with Sta 10 mg/kg obviously inhibited the changes of T wave induced by Iso.Using Jin's formula to calculate the value of q between 0.85 and 2.0,effects of these combination on the changes of T wave were simple combined or enhanced.They obviously reduced the contents of MDA and the activities of LDH in serum,in comparison with Leo 5 mg/kg or Sta 10 mg/kg alone,the combination of Leo 5 mg/kg and Sta 10 mg/kg was remarkably less in the contents of MDA and the activities of LDH.Compared with Leo and Sta,they significantly ameliorated myocardial ischemic injury,only few constriction bands of heart muscle fiber were seen.Conclusion There are more effective of than Leo or Sta alone on acute myocardial ischemic.
