摘要
目的探讨肿瘤坏死因子(TNF-α)对宫颈癌Hela细胞增殖、转移和化疗耐药相关基因表达的影响。方法以外源性TNF-α作用于宫颈癌Hela细胞,作用不同时间点(0-24h)取样分析细胞周期蛋白cyclinD1,锌指蛋白A20,钙调神经磷酸酶CaN,基质金属蛋白酶(MMP-1,MMP-9),金属硫蛋白(MT-1,MT-2)等表达的时间-效应。结果TNF-α作用宫颈癌Hela细胞不同时间段(20min,1h,6h,24h),cyclinD1、A20、CaN、MMP-1、MMP-9、MT-1、MT-2 mRNA的表达都有不同程度改变。RT-PCR检测宫颈癌Hela细胞经TNF-α作用后cyclinD1、MMP-1、MT-2 mRNA的表达6小时达高峰24小时回落,A20、CaN、MMP-9、MT-1 mRNA表达1小时达高峰24小时回落。结论TNF-α诱导宫颈癌Hela细胞cyclinD1、A20、CaN、MMP-1、MMP-9、MT-1、MT-2 mRNA的表达具有时效性。TNF-α可能促进宫颈癌Hela细胞增殖、转移及耐药。
Objective To investigate the effects of tumor necrosis factor-α(TNF-α) on prollieratlon,metastasis and drug resistance-related gene of cervical cancer Hela cells. Methods Hela cells were stimulated by exogenous TNF-α at different time points (0-24h), then gene expressions of cyclinD1, A20, calcineurin (CAN), matrix metalloproteinase (MMP-1,MMP-9), metallothionein (MT-1,MT-2) were detected at mRNA level by reverse transcription-polymerase chain reaction (RT-PCR). Results At different time points post-TNF-α stimulation (20m, 1h, 6h, 24h), the expression of cyclinD1, A20, CaN, MMP-1, MMP-9, MT-1, MT-2 mRNA changed in varous degree. After Hela cells were stimulated by exogenous TNF-α, the expression of cyclinD1 ,MMP-1, MT-2 mRNA reached the peak at 6h and fell back at 24h, and the expression of A20, CaN, MMP-9, MT-1 mRNA reached the peak at lh and fell back at 24h. Conclusions TNF-α-induced expressions of cyclinD1, A20, CaN, MMP-1, MMP-9, MT-1 and MT-2 mRNA in Hela cells is time-dependent, which suggests that TNF-α may be able to promote cervical cancer cells proliferation, metastasis and drug resistance.
