摘要
目的:采用ICR小鼠通过三种行为学实验观察乌灵菌的抗焦虑作用。方法:给药组分别口服乌灵菌粉低、中、高三个剂量0.25、0.50和1.00(g/kg),阳性药对照组为丁螺环酮(2mg/kg)和地西泮(1.5mg/kg),对照组给予等量生理盐水。给药7天后开始行为学实验,包括高架十字迷宫实验、爬梯实验和抗戊四唑致(PTZ)惊厥实验。结果:高架十字迷宫观察5min内小鼠进入开臂的次数(OE),发现乌灵胶囊中剂量组与丁螺环酮组均明显增加(P<0.05,P<0.01),在开臂停留时间(OT)分别占进入两臂总次数和在两臂停留总时间的百分比(OE%和OT%),三剂量组与丁螺环酮组均比对照组明显增加(P<0.01)。记录3min小鼠站立次数和爬梯次数,与对照组比,丁螺环酮组的站立次数和爬梯次数均明显减少(P<0.01),而乌灵胶囊低、高组的站立次数亦明显减少(P<0.05,P<0.01),爬梯次数未见统计学意义。在抗PTZ试验中,中剂量乌灵胶囊明显延长了惊厥发作潜伏期和显著降低了惊厥评分(P<0.01)。结论:三种行为学试验均表明,乌灵胶囊有明确的抗焦虑作用。
Objective: To observe anxiolytic effect of Xylaria nigripes through three behavioral tests in ICR mice. Methods: The mice were orally administered with Xylaria nigripes at different doses of 0. 25, 0. 50 and 1.00 (g/kg) respectively. The mice in positive reference group were treated with Buspirone (2mg/kg) or Diazepam (1. 5mg/kg) and saline was given to control group. 7 days after administration, all the mice were subjected to the behavioral tests, including elevated plus -maze, staircase and PTZ induced convulsion test. Results : OE were significantly increased in Xylaria nigripes group ( P 〈 0. 05 ) and Buspirone group ( P 〈 0. 01 ) within 5 min , and OE% and OT% were increased remarkably in three Xylaria nigripes treatment group as well as Buspirone group compared to control group ( P 〈 0. 01 ). Number of rearing and climbing was significantly reduced in Buspirone group compared to control group ( P 〈 0. 01 ), while the frequency of rearing was remarkably reduced in low and high groups ( P 〈 0. 05, P 〈 0. 01 ). The frequency of climbing was not significant. In Xylaria nigripes group, the latency of convulsion seizure was remarkably prolonged, and the score of convulsion was lowered in the anti -PTZ test (P 〈 0. 01 ). Conclusion: Resuhs of behavioral tests showed that Xylaria nigripes exerted a significant anxiolytie effect.
