在用牛肺巨噬细胞构建了cDNA基因文库的基础上,参照人和小白鼠CD14的基因序列设计一对引物,用PCR方法成功地扩增出编码牛CD14的特异性DNA片段,并进行了序列分析。结果表明,牛CD14基因的核苷酸在相同区域内与人CD14的同源性为79%,推导氨基酸序列的同源性为73%;而与小白鼠的核苷酸和氨基酸的同源性分别为75%和71%。无论在核苷酸还是氨基酸水平上,牛CD14基因与人CD14的同源性都要高于与小白鼠的同源性。
On the base of established cDNA gene library of bovine alveolar macrophages, a pair of primers were designed according to the gene sequences of human and murine CD14 antigens, and then the specifical DNA fragment encoding bovine CD14 antigen was successfully amplified and sequenced by means of PCR. The test results showed that the identities of nucleotides and the deduced amino acids were 79% and 73% between cattle and human, and 75% and 71% between cattle and mouse, respectively, which indicated the gene structure of bovine CD14 antigen is more closely related to human's than that of mouse.
